RESUMO

Brackish groundwater is promising for the cultivation of economically important microalgae; however, its effects have been evaluated only after nutrient supplementation. In this study, 100% brackish groundwater was evaluated as a culture medium for Spirulina sp. (BGWS) and Chlorella fusca (BGWC). In addition, the effects of supplementation with 25% of the nutrients from Zarrouk (BGWS25) and BG-11 (BGWC25) culture media were evaluated. BGWS and BGWC increased the concentration (68.1% w w-1) and productivity of carbohydrate (35.3 mg L-1 d-1) in Spirulina sp. and increased the concentration (56.4% w w-1) and productivity (13.5 mg L-1 d-1) of lipids in C. fusca biomass, when compared to that in the respective controls. The use of brackish groundwater as the sole culture medium is an innovative alternative for the economic production of biomass rich in carbohydrates and lipids. This has potential applications for biofuel production.

RESUMO

This study aimed to evaluate the growth and production of biomolecules by Spirulina sp. LEB 18 cultivated in seawater. The seawater was used without nutrient addition (SW0) and supplemented with 100% (SW100), 50% (SW50), and 25% (SW25) nitrogen, phosphorus, iron, and EDTA concentrations that make up the Zarrouk culture medium. When grown in SW0, Spirulina sp. LEB 18 showed maximum biomass concentration (2.17 g L-1) on the 11th d of cultivation and an increase in the carbohydrate content and productivity by 203% and 52%, respectively, when compared to the control culture. This cultivation strategy demonstrated the feasibility of using seawater as an alternative to freshwater in cultures as well as reduced nutritional requirements for biomass and carbohydrate production.

RESUMO

A priority of the industrial applications of microalgae is the reduction of production costs while maximizing algae biomass productivity. The purpose of this study was to carry out a comprehensive evaluation of the effects of pH control on the production of Nannochloropsis gaditana in tubular photobioreactors under external conditions while considering the environmental, biological, and operational parameters of the process. Experiments were carried out in 3.0 m3 tubular photobioreactors under outdoor conditions. The pH values evaluated were 6.0, 7.0, 8.0, 9.0, and 10.0, which were controlled by injecting pure CO2 on-demand. The results have shown that the ideal pH for microalgal growth was 8.0, with higher values of biomass productivity (Pb) (0.16 g L-1 d-1), and CO2 use efficiency ([Formula: see text]) (74.6% w w-1); [Formula: see text]/biomass value obtained at this pH (2.42 [Formula: see text] gbiomass-1) was close to the theoretical value, indicating an adequate CO2 supply. At this pH, the system was more stable and required a lower number of CO2 injections than the other treatments. At pH 6.0, there was a decrease in the Pb and [Formula: see text]; cultures at pH 10.0 exhibited a lower Pb and photosynthetic efficiency as well. These results imply that controlling the pH at an optimum value allows higher CO2 conversions in biomass to be achieved and contributes to the reduction in costs of the microalgae production process.

RESUMO

The use of different types of concentrated supplements on the performance of Nellore calves grazing Urochloa brizantha cv. Marandu in the dry season was evaluated. The experiment was conducted on 24 ha divided into 12 paddocks. Seventy-two calves with initial body weight (BW) 176±14kg for 140 days of experiment were used. Evaluated treatments included: Mineral, Salt+urea (mineral with 30% urea), Protein (supplementation with 45% crude protein (CP) and 46% total digestible nutrients (TDN)); and Protein+energy (with 28% CP and 73% TDN). The Mineral, Salt+urea, and Protein were offered ad libitum, and the Protein+energy 5g/kg BW. Protein+energy showed the highest (P< 0.05) intake (3.66g/kg BW) followed by the Protein (1.61), Salt+urea (0.36), and Mineral (0.32). The two latter supplements were not significantly different (P> 0.05). The highest (P< 0.05) average daily gain (ADG, kg/day) was observed in the Protein+energy (0.074). Average daily gains for the Protein and Salt+urea (0.014 and -0.024, respectively) were not significantly different (P> 0.05), but were significantly higher (P< 0.05) than the Mineral (-0.085). Therefore, during the dry season, to prevent the loss of bovine weight the supplements must supply in addition to minerals, also energy, non-protein nitrogen (NNP), and true protein.(AU)

The aim of this study was to evaluate the interaction between the periodic addition of monoethanolamine (MEA) and CO2 during the cultivation of Chlorella fusca LEB 111. For this purpose, MEA has been added in abiotic assays, followed by fed-batch cultures with that green alga and the absorbent. BG-11 medium shown a higher potential of CO2 absorption with MEA addition, and the bicarbonate was the chemical species of carbon prevailing in the chemical equilibrium. The periodic addition of MEA did not reduce the kinetics of growth, promoted a higher accumulation of DIC (81.4 mg L-1) in the medium and protein (44.0% w w-1) and lipid (30.8% w w-1) concentrations in the biomass of C. fusca LEB 111. Therefore, it was demonstrated that fed-batch culture with MEA increased CO2 fixation and the biomolecule synthesis as proteins and lipids.

RESUMO

The current study aimed to evaluate if the addition of pentoses along with variations in light intensity and photoperiod can stimulate the production of polyhydroxybutyrate (PHB) and other biomolecules by Chlorella fusca LEB 111. The variables evaluated were the addition of xylose and arabinose as sources of organic carbon, different photoperiods (18 h, 12 h and 6 h light) and variations in light intensities (58, 28 and 9 µmolphotons m-2 s-1). The highest PHB accumulation (17.4% w w-1) and protein production (53.2% ww-1) were observed in assays with xylose addition and a photoperiod of 6 h of light provided at 28 and 58 µmolphotons m-2 s-1, respectively. The highest lipid content (24.7% w w-1) was obtained with 18 h of light. The current study contributes to the development of sustainable alternatives for the use of wastes and the production of biomolecules from algae.

RESUMO

CO2 emissions and the large quantity of lignocellulosic waste generated by industrialized nations constitute problems that may affect human health as well as the global economy. The objective of this work was to evaluate the effects of using CO2 and pentoses on the growth, protein profile, carbohydrate content and potential ethanol production by fermentation of Chlorella minutissima biomass. CO2 and pentose supplementation can induce changes in the microalgal protein profile. A biomass production of 1.84g.L-1 and a CO2 biofixation rate of 274.63mg.L-1.d-1 were obtained with the use of 20% (v.v-1) CO2. For cultures with 20% (v.v-1) CO2 and reduced nitrogen, the carbohydrate content was 52.3% (w.w-1), and theoretically, 33.9mL.100g-1 of ethanol can be produced. These results demonstrate that C. minutissima cultured with the combined use of CO2 and pentoses generates a biomass with high bioenergetic potential.

RESUMO

High concentrations of carbon, which is considered a necessary element, are required for microalgal growth. Therefore, the identification of alternative carbon sources available in large quantities is increasingly important. This study evaluated the effects of light variation and pentose addition on the carbohydrate content and protein profile of Chlorella minutissima grown in a raceway photobioreactor. The kinetic parameters, carbohydrate content, and protein profile of Chlorella minutissima and its theoretical potential for ethanol production were estimated. The highest cellular concentrations were obtained with a light intensity of 33.75µmol.m-2.s-1. Arabinose addition combined with a light intensity of 33.75µmol.m-2.s-1 increased the carbohydrate content by 53.8% and theoretically produced 39.1mL·100g-1 ethanol. All of the assays showed that a lower light availability altered the protein profile. The luminous intensity affects xylose and arabinose assimilation and augments the carbohydrate content in C. minutissima, making this microalga appropriate for bioethanol production.

The aim of this study was to estimate the energy requirements of crossbreed ewe lambs in a feedlot fed with increasing levels of concentrate in the diet. 36 ewe lambs were evaluated with initial body weight of 23.7±3.67kg. Six animals were slaughtered at the beginning of the experiment to obtain the reference group values. Animals (30) were distributed in five treatments: maintenance (feed restricted with Tifton grass hay) and those supplemented with increasing dietary levels of concentrate (20, 40, 60 and 80%) on a dry matter basis (six animals per treatment). The ewe lambs were slaughtered when they reached 37.70±9.89kg. The concentrate levels influenced the utilization efficiency of metabolizable energy for maintenance (km) and consequently the daily requirements of metabolizable energy for maintenance. The net energy requirements for maintenance of crossbred lambs in growth is 57kcal/FCW 0.75/day, where FCW is body weight on fasting. The ewe lambs with 20kg presented net energy requirement for 100g daily weight gain of 465kcal/day. The ewe lambs with 40kg of body weight required 930 kcal/day of net energy for the same gain. The net energy requirement for maintenance of ewe lambs can be estimated by the equation: NEm (Mcal/day) = 0.057 * FCW0.75. The net energy requirement for crossbred ewe lambs gain (Mcal/day) can be estimated from the equation: NEg (Mcal/day) = 0.524*FCW 0.75*GEBW1.21, where GEBW represented gain empty body weight. Diets affect the efficiencies of uses of metabolizable energy for maintenance (km) and gain (kg).(AU)

RESUMO

This study aimed to examine the metabolic changes in Chlorella minutissima cells grown under nitrogen-deficient conditions and with the addition of xylose. The cell density, maximum photochemical efficiency, and chlorophyll and lipid levels were measured. The expression of two photosynthetic proteins, ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) and the beta subunit (AtpB) of adenosine triphosphate synthase, were measured. Comparison of cells grown in medium with a 50% reduction in the nitrogen concentration versus the traditional medium solution revealed that the cells grown under nitrogen-deficient conditions exhibited an increased growth rate, higher maximum cell density (12.7×10(6)cellsmL(-1)), optimal PSII efficiency (0.69) and decreased lipid level (25.08%). This study has taken the first steps toward protein detection in Chlorella minutissima, and the results can be used to optimize the culturing of other microalgae.

RESUMO

The aim of this study was to evaluate the effect of increasing levels of concentrate on the quantitative and qualitative carcass characteristics of feedlot ewes. Twenty-four crossbred ewes were randomly distributed in four treatments: (1) diet with 20%; (2) 40%; (3) 60%; and (4) 80% of concentrate. Animals were slaughtered when those receiving the diet with 80% of concentrate reached adult weight, at around 50kg LW. The data were analyzed by means of variance and regression analyses and correlations. The levels of concentrate increased live weight at slaughter linearly. Height at the withers and rump presented medium to high correlation (0.86). The correlation of loin eye area measured on the carcass and by ultrasound examination was 0.81, whereas for back fat thickness it was 0.85. The increase in the use of concentrate in the diets promotes heavier carcasses with better yields and fat cover. Some measures obtained in vivo presented high correlation with those measured on the carcass. The use of ultrasound can be a way to predict the in vivo loin eye area. The use of concentrate improves the meat quality in variables' color and appearance...

The association of stem cells (SCs) with biomaterials promises to be the protagonist for future regenerative medicine in the treatment of tissue and organ lesions. Stem cells were cultivated in scaffolds constructed by the electrospinning technique, using poly-D,L-lactic acid (PDLLA) associated or not with Spirulina biomass (PDLLA/Sp), which has bioactive components of interest for tissue engineering (TE). Physicochemical analyses were performed, such as morphology, fiber diameter, degradability, residual solvent, roughness, contact angle with water, among others. SCs adhesion, proliferation and scaffold cytotoxicity were also evaluated. Nanofibers without beads and with characteristics similar to the natural extracellular matrix (ECM) in terms of mechanical and topographical properties were obtained. In biological tests it was found that SCs adhered more and had greater viability in the PDLLA/Sp molds, when compared with the PDLLA scaffolds. The scaffolds were shown to be atoxic for the SCs. It can be concluded that the scaffolds developed in this work have the characteristics to be a new biomaterial suitable for use in TE.

The influence of concentrate levels in rations on ingestion behavior, nutrient intake, and digestibility was determined in cattle. Four 442.15kg BW crossbred cows were distributed in a 4x4 Latin square, into four periods of 14 days, 11 days for adaptation and three days for sample collections per animal and per period. Behavior was determined by observing each animal every 15 minutes for 24 hours, recording feeding, rumination, idling, and water uptake time. The roughage was whole-plant corn silage and the concentrate was composed of corn, sorghum, soybean meal and hulls, minerals, urea, and protected fat, in rations with 13 percent crude protein. The concentrate levels were 30, 40, 50, and 60 percent in dry matter. Intake and rumination activities decreased linearly with the concentrate level, and consequently, there was increasing idling time. A linear effect was observed for concentrate levels on intake and rumination efficiencies, as well as for intake of the dietary nutrients, but not for neutral detergent fiber intake. Increases in the concentrate level decreased feeding and rumination times, but did not alter nutrient digestibility in the diet.

RESUMO

We evaluated the kinetic culture characteristics of the microalgae Cyanobium sp. grown in vertical tubular photobioreactor in semicontinuous mode. Cultivation was carried out in vertical tubular photobioreactor for 2 L, in 57 d, at 30 °C, 3200 Lux, and 12 h light/dark photoperiod. The maximum specific growth rate was found as 0.127 d(-1), when the culture had blend concentration of 1.0 g L(-1), renewal rate of 50%, and sodium bicarbonate concentration of 1.0 g L(-1). The maximum values of productivity (0.071 g L(-1) d(-1)) and number of cycles (10) were observed in blend concentration of 1.0 g L(-1), renewal rate of 30%, and bicarbonate concentration of 1.0 g L(-1). The results showed the potential of semicontinuous cultivation of Cyanobium sp. in closed tubular bioreactor, combining factors such as blend concentration, renewal rate, and sodium bicarbonate concentration.

The influence of concentrate levels in the diet of cattle on intake, rumen pH, and ammonia nitrogen (NH3-N) using four cows fistulated in the rumen and distributed in 4x4 Latin square was evaluated. Corn silage was used as roughage and concentrate was maize, sorghum, soybean hulls, soybean rules, urea, and protected fat. Diets were isonitrogenous with 18.6 percent crude protein, using increasing amounts of ether extract (5.7, 6.6, 7.5, and 8.4 percent) to the levels of concentrate, 30, 40, 50 and 60 percent. The average values of rumen pH were 6.71, 6.64, 6.63, and 6.69 and the average concentrations of NH3-N were 37.40, 39.62, 35.59, and 34.13mg/100mL of rumen fluid. Animals fed 7.5 percent ether extract diet had higher neutral detergent fiber intake than the animals of other treatments with maximum dry matter intake estimated in 10.47kg/day. Protected fat diet did not interfere in the environment of rumen up to levels of 8.4 percent of ether extract, providing better efficiency with respect to intake of 55 percent concentrate.

RESUMO

BACKGROUND AND PURPOSE: The influence of apolipoprotein E (ApoE) polymorphism on clinical severity of multiple sclerosis (MS) is still controversial. Cigarette smoking has been suggested to influence the progression of disability in these patients. In this study, we aimed to investigate whether an interaction of smoking with the ApoE polymorphism influences the progression of disability in MS patients. METHODS: Smoking history from 205 female patients with MS was obtained. Clinical data collected include age at onset, disease duration, annual relapse rate, the Expanded Disability Status Scale (EDSS) and the Multiple Sclerosis Severity Score (MSSS). ApoE polymorphism was examined in all patients and stratified according to smoking status and associations with the clinical data investigated. RESULTS: There were no significant associations between cigarette smoking and any of the clinical characteristics in the whole group of patients. In women carrying the ApoE E4 isoform, smokers had a lower EDSS (P = 0.033) and MSSS (P = 0.023) in comparison with non-smokers. CONCLUSION: Our data suggest that in women with MS carrying the ApoE E4 isoform, cigarette smoking may have a protective influence on disease progression and accumulation of disability. These findings need to be confirmed by future large longitudinal studies.

The intake, the digestibility of nutrients, and the total digestible nutrients (TDN) content in sheep diets based on corn and sorghum grains high moisture silages with or without microbial inoculant, were evaluated. The used roughage was Brachiaria hay (Brachiaria brizantha cv. Marandu). Twelve animals were distribuited to treatments: (1) hay+high moisture corn grain silage, (2) hay+high moisture sorghum grain silage; (3) hay+high moisture corn grain silage with microbial inoculant, and (4) hay+high moisture sorghum grain silage with microbial inoculant. No differences among treatments in relation to the intake of dry matter, organic matter, crude protein, neutral detergent fiber, and total and non-structural carbohydrates were observed. The ether extract intake differed according to the grains. The silages did not differ in relation to the estimated digestibilities for dry matter, organic matter, crude protein, neutral detergent fiber, ether extract, and total and non-structural carbohydrates. The TDN did not differ according to the grains and inoculant, with averages of 77.6 and 79.5 percent, for corn and sorghum grains high moisture silages, respectively. It was demonstrated that the use of microbial inoculant, in high moisture grain silages was not necessary. High moisture corn grain silage can be substituted by high moisture sorghum grain silage without effects on the use of nutrients by sheep.

The effect of the castration on the productive performance and carcass characteristics was studied, as well as, the economic evaluation of finished bovines raised on Brachiaria decumbens pastures. Eight Canchim-Nellore steers aging 14-months-old were used, being four castrated and four non-castrated, averaging 273.2kg body weight. It was used a completely randomized design with four repetitions per treatment. The animals received 0.7 percent of their body weight of proteic-energetic supplement and were slaughtered at 26 months of age. The weight at slaughtering and the average daily weight gain differed between castrated and non-castrated, averaging 441.0 and 482.2kg, and 0.6 and 0.7kg/day, respectively. No effects of the castration on carcass characteristics were observed, except for weight of hot carcass, 252.3 versus 229.9kg; meat color, 3.25 versus 4.50 points; and subcutaneous fat thickness, 0.6 versus 1.4mm; respectively, for non-castrated and castrated. The profitabilities per animal and hectare were 14.5 and 15.8 percent for non-castrated and 4.5 percent and 5.8 percent for castrated, respectively. It is suggested the raised of non-castrated bovines supplemented on Brachiaria decumbens pastures.

RESUMO

OBJECTIVE: To assess the effects of fish (lean or oily) and fish oil consumption on blood lipid concentration during weight loss. DESIGN: Randomized, controlled 8-week trial of energy-restricted diet varying in fish and fish oil content. Subjects, 324 men and women, aged 20-40 years, body mass index 27.5-32.5 kg m(-2), from Iceland, Spain and Ireland, were randomized to one of four groups: (1) control (sunflower oil capsules, no seafood), (2) cod diet (3 x 150 g week(-1)), (3) salmon diet (3 x 150 g week(-1)), (4) fish oil (DHA/EPA capsules, no seafood). The macronutrient composition of the diets was similar between the groups and the capsule groups were single-blinded. MEASUREMENTS: Total cholesterol (TC), high-density lipoprotein (HDL) and low-density lipoprotein cholesterol, triacylglycerol (TG) and anthropometrics were measured at baseline and end point. RESULTS: The difference in logTG lowering between the control group and the cod diet, salmon diet and fish oil from baseline to end point was -0.036 (95% CI -0.079 to 0.006), -0.060 (-0.101 to -0.018) and -0.037 (-0.079 to 0.006), respectively. Reduction in TC was about 0.2 mmol l(-1) greater in the fish groups (cod and salmon) than in the control group, but only of borderline significance when adjusting for weight loss. HDL tended to decrease less in the diet groups consuming a significant amount of n-3 fatty acids (salmon and fish oil). CONCLUSION: Weight-loss diet including oily fish resulted in greater TG reduction than did a diet without fish or fish oil. Controlled trials using whole fish as a test meal are encouraged to be able to elucidate the role of different constituents of fish for human health.

Performances of cows and calves weaned at 90 (D90) and 210 days of age (D210) were evaluated, using 36 Beefalo-Nelore multiparus cows and their calves, in a randomized block experimental design, with 18 replicates of nine animals of each sex. Calves were allotted to treatments using as criteria to form blocks, the weight and sex of the animal. Calves (D210) were raised in 48ha pasture of Brachiaria decumbens e B. humidicula. Calves (D90) were raised in feedlot condition, in an area of 7.95m² /animal, and were fed chopped elephant grass and concentrate. The average concentrate dry matter intake, weaned body weight and weight gain were, respectively, 0.56 and 1.69kg/day; 208.68 and 185.67kg; 0.75 and 0.52kg/day respectively for D210 e D90 calves. Cows from D90 treatment showed higher body weight (448.86kg) at 210 days post partum. Creep-feeding is suggested for weaning heavier calves
